Radioactivity is physically introduced into the sample by adding a measured amount of a radioactive species called a tracer.

A weighed quantity of radioactively tagged analyte having a known activity is added to a measured amount of the sample.  After thorough mixing to ensure homogeneity, a fraction of the component of interest is isolated and purified.  The activity of the isolated fraction is then measured and used to calculate the mass of the component of interest in the original sample.
